During myocardial remodeling, extracellular matrix components are altered. Among them, laminin is a critical component of the basement membrane in the heart. The cardiac laminin receptor is a transmembrane protein with two extracellular domains. Its cytoplasmic domain binds to tyrosine kinase receptors and leads to signal transduction. Little is known about the expression of the secreted form of this receptor in the heart. We assessed the differential expression of this soluble form in the myocardium. Myocardial remodeling was induced by coronary artery ligation in male rats. After 14 d, the secreted form of the 130-kDa laminin receptor was found to be increased in noninfarcted myocardial regions, and this increase was accompanied by the presence of extra matrix deposited in the infarcted area. After 26 d, the level of the 130-kDa secreted laminin receptor was higher in the noninfarcted region compared with that at 14 d, suggesting an ongoing effect of the ligation. As shown by immunohistochemistry, the increase of the 130-kDa secreted laminin receptor was mainly associated with the nonmyocytes, and in the infarct area, it was mainly detected in the myocardial capillaries. In contrast, no significant change in the expression of the 120-kDa membrane laminin receptor was observed. These findings may be important in understanding the different pathophysiological processes underlying fibrosis and inflammation in the infarcted myocardium and in identifying the laminin receptor as a potential pharmacological target.Aiden James
